1.PENDAHULUAN
1.1. Latar Belakang
Sistem-sistem manajemen keamanan pangan berdasarkan ISO 22000 dapat membantu organisasi untuk mengurangi risiko-risiko yang berkaitan dengan makanan dan minuman. Sistem-sistem manajemen ini juga tidak hanya memperhitungkan aturan dasar dalam membuat makanan dan praktek-praktek tempat kerja yang dapat diterima secara benar, tetapi juga meliputi rencana-rencana yang memungkinkan untuk terjadinya kesalahan dalam pengolahan sehingga dimungkinkan untuk penarikan kembali produk. Semua jenis praktek tersebut membentuk dasar suatu sistem manajemen keamanan pangan. Standar ini mencakup key elements untuk membentuk keamanan pangan, dimana salah satu key element tersebut adalah HACCP yang dirancang untuk digunakan pada semua segmen industri pangan mulai penanaman, pemanenan, pengolahan, pabrikasi, distribusi dan penjualan sampai pada penyiapan makanan untuk dikonsumsi. Program-program prasyarat seperti GMP yang diterapkan saat ini (current Good Manufacturing Practices) merupakan suatu dasar yang yang penting bagi keberhasilan pengembangan dan penerapan rencana HACCP. Sistem keamanan pangan yang didasarkan pada HACCP telah diterapkan dengan sukses pada pabrik pengolahan makanan, toko penjual makanan dan operasi jasa pelayanan makanan (Kurniawan, 2008).
Kebutuhan akan keamanan pangan mulai dari semua organisasi yang menghasilkan, membuat, menangani atau menyediakan makanan merupakan kebutuhan yang tertinggi. Lagipula, semua organisasi ini harus mengenali secara terus menerus untuk meningkatkan kebutuhan untuk mempertunjukkan dan menyediakan cukup bukti dari kemampuan mereka untuk mengidentifikasi dan mengendalikan bahaya dari keamanan pangan dan banyak kondisi-kondisi yang berdampak pada keamanan pangan. Kebutuhan ini ditujukan untuk semua jenis dalam rantai makanan yang berkisar antara produsen ternak, produsen utama melalui pembuatan makanan, transpotasi dan operator gudang dan subkontraktor untuk eceran dan pelayanan distribusi makanan bersama-sama dengan organisasi lain seperti perlengkapan, bahan pengemasan, agen pembersih, ramuan dan bahan tambahan (Anonima, 2006).
Sistem HACCP ada tujuh prinsip :
-Analisa hazard yang meliputi penilaian dan identifikasi ancaman dan penentuan tentang hazard dan ukuran control hazard dan metoda dalam menetralkan ancaman hazard.
-Penentuan CCP (Critical Control Point).
-Penetapan titik kritis dari identifikasi CCP.
-Penentuan dan implementasi dari sebuah sistem dengan monitoring dari CCP.
-Penetapan dari aksi koreksi.
-Penetapan prosedur verifikasi dalam rangka mengkonfirmasikan jika sistem ini efektif dan bertindak sesuai rencana.
-Menetapkan dokumentasi mengenai semua prosedur dan catatan yang sesuai dengan prinsip-prinsip dan penerapannya.
2. APLIKASI ISO DAN TURUNAN ISO 22000
HACCP (Hazard Analysis Critical Central Point) atau ISO 22000 mengenai sistem manajemen keamanan pangan adalah proses sistem kontrol yang di desain untuk identifikasi dan mencegah mikrobia dan bahaya lainnya dalam produksi makanan dan keseluruhan rantai makanan. HACCP meliputi tahapan pendesainan untuk mencegah masalah sebelum hal ini terjadi dan untuk mengoreksi penyimpangan secara sistematis secara cepat dapat mendeteksi masalah yang ada. HACCP/ISO 22000 memungkinkan produsen, pengolah, distributor, eksportir dan lain sebagainya dari produk pangan untuk menggunakan sumber daya teknik secara efisien dan dalam cara biaya yang efektif dalam jaminan keamanan pangan (Anonima, 2005).
Susunan jaminan mutu paling banyak didasarkan pada prinsip manajemen mutu dari ISO 9000/ISO 22000 dan konsep HACCP, dalam penambahan beberapa penyusunan GAP (Good Agricultural Practice). Dasar standar mutu menyediakan perbaikan dari kejelasan proses, membantu untuk mendeteksi dan menghindari kegagalan sistematik dan sebuah kesempatan yang lebih baik untuk traceability. ISO 22000 adalah perbaruan dari standard ISO 9000 dan juga yang mengkombinasikan standar ISO 9000 dan konsep HACCP ke dalam satu standar. Bagaimanapun, perbedaan yang utama antara ISO 22000 dan ISO 9000 mengenai ruang lingkupnya. Pertama dengan tujuan keamanan pangan, sedangkan yang lainnya mengarahkan pada mutu pangan. Standar ISO 22000 dimaksud untuk menjadi bagian yang independen dan dapat digunakan untuk semua jenis organisasi di dalam penyedia rantai makanan. HACCP digunakan untuk industri pangan dan tujuan untuk menetapkan produksi yang baik, sanitasi dan manufaktur untuk menghasilkan pangan yang aman dan untuk pro aktif dan pencegahan lebih baik daripada menimbulkan reaksi. Konsep dari HACCP dapat diterapkan pada semua tahapan dari sistem pangan. Implementasi dari HACCP dan GMP kepada seluruh rantai yang termotivasi oleh faktor internal dan eksternal, seperti peningkatan efisiensi yang internal dan akses pasar sebagai faktor eksternal (Gellynck & Kühne, 2007).
ISO 22000 adalah standar internasional yang menggambarkan kebutuhan dari suatu sistem manajemen keamanan pangan yang mencakup semua organisasi dalam rantai makanan dari "farm to fork". Kombinasi standar umumnya mengetahui unsur-unsur kunci untuk menentukan keamanan pangan sepanjang rantai makanan, yang meliputi:
-Komunikasi interaktif.
-Sistem manajemen.
-Pengendalian dari bahaya keamanan pangan ke arah persyaratan penuh dari program dan perencanaan HACCP.
-Peningkatan yang berkelanjutan dan pembaharuan dari sistem manajemen keamanan pangan(Anonimb, 2005).
Hazard Analysis Critical Control Point (HACCP) menjamin dari segi keamanannya sedangkan ISO 9001 lebih fokus dalam menjamin kualitas produk. Dengan mengaplikasikan HACCP dengan ISO 9001 quality management system menghasilkan sistem yang lebih efektif daripada hanya menggunakan HACCP atau ISO 9001 secara sendiri-sendiri. Hal ini juga bertujuan untuk meningkatkan kepuasan konsumen dan memperbaiki keefektifan dalam pengorganisasiannya (Sparling et al., 2001 dalam Hernández et al, 2003). Hal tersebut tercakup apabila mengiplementasikan ISO 22000 dalam pelaksanaan proses. ISO 22000 lebih konsentrasi pada keamanan pangan dan prosedur instruksi bagaimana membangun sistem keamanan pangan tersebut. (Petro-Turza, 2003 dalam Hernández et al, 2003). Pada tahun 2001, dalam rangka untuk memudahkan implementasi HACCP dan ISO 9001 dalam organisasi pangan, maka organisasi intemasional dengan standardisasi (ISO) telah menerbitkan petunjuk tentang aplikasi dari ISO 9001:2000 untuk industri makanan dan minuman (ISO 15161:2000). Petunjuk ini dapat memberi perkiraan mengenai bagaimana ISO 9001 bisa diterapkan pada organisasi pangan dan yang dirancang untuk organisasi yang melibatkan dalam semua aspek industri makanan. ISO 15161:2000 meliputi permulaan, memprosesan dan pengemasan produk makanan dan minuman dan menjelaskan kemungkinan untuk menghubungkan komunikasi antara kedua system tersebut dan penggunaan dari komponen yang sama. Ini penting untuk mempertimbangkan, ISO 15161 yang bukan merupakan standar HACCP dan tidak bisa digunakan sebagai acuan pada sertifikasi dokumen, tetapi petunjuk ini yang diharapkan untuk menyediakan sistem manajemen yang bersih yang mendukung pada pengendalian HACCP untuk sistem keamanan pangan yang efektif, diketahui dibawah kerangka dari ISO 9000 tentang Sistem Manajemen Mutu. Pada sisi lain, ISO 22000 berkonsentrasi secara eksklusif pada keamanan pangan dan akan diinstruksikan pada produsen makanan bagaimana mereka dapat membangun sistem keamanan pangan secara mandiri.
2.1. Manfaat dan Kendala Penerapan ISO 22000
Penerapan ISO 22000 bermanfaat untuk membantu kita dalam mencapai beberapa sasaran, yang meliputi:
-Penetapan sistem manajemen keamanan pangan (FSMS).
o Untuk perencanaan dan implementasi FSMS dari organisasi kita.
o Untuk pengoperasian dan memelihara organisasi FSMS.
o Untuk memperbaharui dan meningkatkan organisasi FSMS.
-Untuk memastikan bahwa produk tidak menyebabkan efek kesehatan yang kurang baik.
-Untuk menunjukkan penyesuaian dengan kebutuhan keamanan eksternal.
o Untuk menujukkan penyesuaian dengan kebutuhan keamanan yang legal.
a. Untuk menunjukkan penyesuaian dengan kebutuhan regulasi.
b. Untuk menunjukkan penyesuaian dengan kebutuhan yang menurut hukum.
o Untuk menunjukkan penyesuaian kebutuhan konsumen.
- Untuk mengevaluasi keamanan pangan dari konsumen kita.
- Untuk menyediakan produk yang aman dan meningkatkan kepuasan pelanggan.
- Untuk produk pangan ekspor dan menembus pasar internasional.
- Untuk isu komunikasi keamanan keseluruh rantai makanan.
o Untuk komunikasi dengan organisasi pelanggan kita.
o Untuk komunikasi dengan organisasi penyalur kita.
o Untuk komunikasi kepada pihak-pihak lain yang relevan.
- Untuk memastikan bahwa kita mematuhi kebijakan keamanan pangan.
o Untuk menunjukkan penyesuaian untuk semua pihak yang berkepentingan.

Pada beberapa negara maju dan berkembang termasuk Indonesia dalam menerapkan sistem HACCP mengalami kendala dalam penerapannya terutama pada usaha kecil. Kendala yang dihadapi usaha kecil, seperti sumber keuangan, keahlian manajemen dan teknis. Sedangkan pada usaha katering hambatannya adalah pengetahuan, pelatihan, petinggi staf, variasi produk yang besar, variasi dalam permintaan dan beban kerja, dan banyaknya pekerja paruh waktu (Murat et al., 2007).
2.2. Turunan ISO 22000
- ISO/TS 22004, sistem manajemen keamanan pangan: mengarah kepada aplikasi dari ISO 22000:2005, yang dipublikasikan bulan November 2005, yang menyediakan bimbingan penting yang dapat membantu organisasi yang mencakup perusahaan sedang dan menengah yang ada diseluruh dunia.
- ISO/TS 22003, sistem manajemen keamanan pangan: merupakan kebutuhan dari asal badan audit dan sertifikasi dari sistem manajemen keamanan pangan, akan memberi bimbingan yang seimbang pada akreditasi (penerimaan) tentang ISO 22000 dengan badan sertifikasi dan menggambarkan aturan untuk pengauditan sistem manajemen keamanan pangan ketika menyesuaikan diri kepada standar ini. Dan akan diterbitkan dalam kwartal pertama tahun 2006.
- ISO 22005, penerapan treaceability dalam makanan ternak dan rantai makanan: prinsip umum dan bimbingan dari desain sistem dan pengembangan, akan segera dikeluarkan sebagai draf standar internasional.

2.2.a. ISO 22003 sistem manajemen keamanan pangan
ISO/TS 22003:2007 akan membantu untuk menciptakan kepercayaan dalam sertifikasi keseluruh dalam persediaan rantai makanan. ISO/TS 22003 merupakan dokumen yang terakhir dalam rangkaian ISO untuk sistem manajemen keamanan pangan, yang menyeimbangkan kelayakan keamanan pangan dalam prakteknya di seluruh dunia. Ini diluncurkan pada tahun 2005 dengan ISO 22000, yang didukung oleh suatu konsensus internasional antar tenaga ahli dari pemerintah dan industri (Anonimb, 2007).
2.2.b. ISO 2005 penerapan traceability dalam makanan ternak dan rantai makanan
Standarisasi ini memperbolehkan pengoperasian pada tiap tahapan dari rantai makanan untuk :
- Melacak alir bahan (makanan ternak, makanan, ramuan dan pengemasan mereka).
- Mengidentifikasi keperluan dokumentasi dan pelacakan dari masing-masing langkah dari produksi.
- Memastikan koordinasi yang cukup antara para pemeran yang dilibatkan secara berbeda.
- Membutuhkan masing-masing pihak yang diinformasikan langsung dari penyalur yang paling sedikit dan pelanggan dan lain sebagainya.
Sebuah sistem traceability memperbolehkan organisasi untuk membuat dokumen dan atau lokasi produk melalui tahapan dan dioperasikan yang dilibatkan dalam manufaktur, pemprosesan, distribusi, dan penanganan dari makanan ternak dan makanan, dari produk utama ke konsumen. Oleh sebab itu mendapat fasilitas untuk identifikasi penyebab dari tidak sesuaian dari produk, dan kemampuan untuk menggambarkan dan atau menginggat kembali itu dibutuhkan (Anonima, 2007).
2.3. Penerapan aplikasi ISO 22000
ISO 22000 dapat digunakan oleh:

- Pengolahan daging.

- Pabrikan roti.
- Pabrikan gandum.
- Pembalut luka pabrikan.
- Pabrikan hidangan.
- Pabrikan bumbu.
- pabrikan pengemasan.
- Pabrikan makanan yang dibekukan.
- Pabrikan makanan kalengan.
- Pabrikan manisan.
- Pabrikan tambahan aturan makanan.

2.4. Contoh Implementasi ISO 22000
Timbulnya berbagai penyakit dan kontaminasi pada produk pangan yang berasal dari hewan. Hal ini dapat menimbulkan kecemasan masyarakat yang ada di seluruh dunia, yang pada akibatnya berpengaruh kepada sikap konsumen. Oleh sebab itu konsumen menjadi ingin tahu informasi lebih banyak mengenai sumber produk yang mereka konsumsi. Karena adanya tekanan dari konsumen maka banyak perubahan yang terjadi di dalam produksi makanan, baik dari segi kualitas maupun keamanannya
Dalam penerapan untuk mendapatkan kualitas dan keamanan yang efektif, maka perusahaan mulai memperhatikan kepentingan konsumen. Lawton (2002) dalam Hernández et al (2003) menggambarkan prioritas dari konsumen yang diawali dengan membangun penilaian yang berkaitan dengan proses, produk dan hasil. Proses pengerjaan dikaitkan dengan karakteristik yang diinginkan konsumen.
Dalam kurun waktu 10 tahun belakang ini, telah banyak dijumpai kejadian yang berkaitan dengan keamanan pangan dan kualitas pangan salah satunya adalah dalam industri daging. Kejadian tersebut menyebabkan suatu industri harus dievaluasi kembali mengenai bagaimana supply chain daging dan tentang kerusakan yang diakibatkan penanganan saat berada di pasar.
Beberapa penelitian tentang industri daging di Eropa Barat telah dilakukan oleh banyak peneliti yang diantaranya adalah Fearne(1998); Viaene dan Verbeke (1998); Lobstein (2001); Yeung dan Morris (2001) dalam Goldsmith1, et al (2002) yang menggambarkan perubahan secara drastis yang disebabkan adanya ketakutan pada produk pangan dan ketidakpercayaan masyarakat terhadap sistem. Kejadian Bovine Spongiform Encephalopathy (BSE) dan kembali munculnya penyakit kaki dan mulut (Foot and Mouth Disease / FMD) serta adanya kontaminasi biologi dan kimia pada produk makanan yang menggoncang sejumlah industri daging di Eropa Lobstein (2001) dalam Goldsmith1, et al (2002) menyarankan bahwa industri daging seharusnya tidak hanya menerima reaksi atas permasalahan ini tetapi juga seharusnya telah menyiapkan diri dalam menghadapi permasalahan keamanan pangan. Dailey (2001) dalam Goldsmith1, et al (2002) juga mengatakan bahwa perlu adanya suatu sistem yang bisa dipercaya dalam menjamin mutu yang dihasilkan untuk industri daging. Martin (2001) dalam Goldsmith1, et al (2002) juga menyampaikan, berdasarkan survei Food Marketing Institute di USA dikatakan bahwa kepercayaan konsumen di USA berkaitan mutu pangan mulai terkikis dari tingkat kepercayan 84% (tahun 1996) menurun menjadi 74% (tahun 2000). Berkaitan dengan penelitian tersebut itulah maka dikatakan bahwa ketakutan akan pangan dan tuntutan sosial dapat menjadi sumber yang berasal dari luar, yang bisa mendorong industri memulai proses perubahan fundamental seperti yang terjadi di Eropa.
Penilaian kehigenisan daging dengan menyediakan suatu pendekatan pemonitoran yang distandarisasi yang mana staf perusahaan dilatih, untuk tujuan mengukur ketepatan dari HACCP yang didasarkan pada proses pengendalian MSQA, pada langkah-langkah yang spesifik pada garis pengolahan. Tindakan korektif diperlukan untuk sistem pelaksanaan yang tidak cukup, sebagai contoh, melalui SOPs atau instruksi kerja jika dibutuhkan. Sistem MHA dapat membantu untuk memastikan keseragaman hasil pengecekan dari pengolahan industri daging ekspor di Australia, dan mencerminkan kebutuhan akan ketidakadaan toleransi dari kenyataan faecal dan pencemaran ingesta pada daging (Buttler et al., 2003).
3. KESIMPULAN
-Sistem-sistem manajemen keamanan pangan berdasarkan ISO 22000 dapat membantu organisasi untuk mengurangi risiko-risiko yang berkaitan dengan makanan dan minuman.
-Sistem keamanan pangan yang didasarkan pada HACCP telah diterapkan dengan sukses pada pabrik pengolahan makanan, toko penjual makanan dan operasi jasa pelayanan makanan.
-Keamanan pangan dihubungkan dengan keadaan dan tingkat bahaya kerusakan pangan pada poin konsumsi.
-Susunan jaminan mutu paling banyak didasarkan pada prinsip manajemen mutu dari ISO 9000/ISO 22000 dan konsep HACCP.
-Dalam menerapkan ISO 22000 selain memperoleh keuntungan ternyata para pengusaha juga menemui kendala.
-Varian dari ISO 22000 adalah ISO 22003, 22004 dan 22005.
-ISO 22000 dapat diterapkan pada semua bidang.

DEFINITION, MECHANISMS, FUNCTIONAL OF OMEGA-3 FATTY ACID
1. INTRODUCTION
Omega-3 fatty acids are being increasingly promoted as important dietary components for health and disease prevention. These fatty acids are naturally enriched in fatty fish like salmon and tuna and in fish-oil supplements. An increasing number of foods that are not traditional sources of omega-3 fatty acids, such as dairy and bakery products, are now being fortified with small amounts of these fatty acids. Fatty acids have many fates in the body, including β-oxidation for energy, storage in depot fat or incorporation into phospholipids, which form the major structural components of all cellular membranes Because humans do not have the enzymatic machinery required to synthesize omega-3 fatty acids, they must be obtained from the diet (termed “essential fatty acids”) (Suretter, 2008).
Omega-3 fatty acids are a family of fatty acids that contain two or more double bonds (“polyunsaturated”). One of which is located three carbon positions from the methyl terminus (“omega-3” or “n-3”) because the body is unable to synthesize in appreciable amounts omega-3 fatty acids that are longer than 14 carbons they are obtained primarily from dietary sources (Friedman & Moe, 2006).
Fish such as tuna, trout and salmon are especially rich sources of these fatty acids. Fish oil supplements are also a rich source as they typically contain 30%–50% omega-3 fatty acids by weight. Small quantities of omega-3 fatty acids are naturally present in meats like beef, pork and poultry. But also is plant sources such as flax and canola oil. The marine omega-3 fatty acids preparation we used contains a small amount of antioxidant vitamin E (equal to approximately 16 IU vitaminE/day), which theoretically may be expected to have an effect. (Surette, 2008 ; Goodfellow et al., 2009).
The phospholipid polyunsaturated fatty acids (PUFAs) hypothesis of depression is enlightening a promising path to discover the unsolved of depression. There are two main types of PUFAs in the human body. The omega–6 (n–6) series derived from cis-linoleic acid (LA, 18: 2) and the omega–3 (n–3) series derived from α-linolenic acid (ALA, 18: 3), n–3 and n–6 PUFAs are important constituents of all cell membranes. They are essential for survival of humans and other mammals, and cannot be synthesized in the body. The inability of vertebrates to synthesis linoleic acid (18:2ω6) and α-linolenic acid (18:3ω3) leads to essential requirement for these fatty acids (FAs) in diet (Pinsu, 2009 ; Dennis et al., 1993). There are three major types of omega 3 fatty acids including alpha-linolenic acid (ALA), eicosapentaenoic acid (EPA), and also docosahexaenoic acid (DHA) (Alshatwi & Alrefai, 2007). Omega-3 fatty acids such as eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA) are highly polyunsaturated and readily undergo oxidation (Sethi et al., 2002).
Omega-3 polyunsaturated fatty acids (PUFAs) are one of dietary substrates that have reported cardioprotective benefits. Omega-3 PUFAs generally exert their cardioprotective effects through changes in lipids and lipoproteins (Alshatwi & Alrefai, 2007). The structural role of long chain polyunsaturated fatty acids (LCPUFAs) and the functional correlates of specific Fas are being increasingly recognized. The LCPUFAs arachidonic acid (20:4w6) is an important membrane component and precursor to prostaglandins, leucotrienes, and epoxides (Dennis et al., 1993).
The aim of this study was to determine the effects of omega-3 fatty acid in human health. And known mechanisme from work omega-3 fatty acid.
2. MEKANISME OF OMEGA-3
The biological mechanisms to explain the role of n–3 fatty acids in depression are the regulation of neurotransmitters and signal transduction by PUFAs. The change in fatty acid concentration in the brain, induced by chronic deficiency in dietary n–3 fatty acids, could lead to an increase in serotonin 2 (5-HT2) and decrease in dopamine 2 receptor density in the frontal cortex. The upregulation of 5-HT2A/C receptors and down regulation of dopamine receptor are thought to play a role in the pathophysiology of depression. Furthermore, high cerebrospinal fluid concentration of 5-hydroxy-indoleacetic acid (5-HIAA), a metabolite of serotonin and an indicator of brain serotonin turnover, has been shown to be associated with high plasma concentration of n–3 PUFAs among healthy human.
The other possible biological mechanisms of the beneficial effects of n–3 PUFAs on mood and physical illness are regulation of the corticotropin-releasing factor, inhibition of protein kinase C, suppression of phosphatidylinositol-associated second messenger activity, modulation of heart rate variability via parasympathetic nervous system, increased dendritic arborization and synapse formation, promotion of neuroprotection and prevention of neuronal apoptosis, and synthesis of neuroprotectin D1 inhibit angiotensin-converting enzyme and 3-hydroxy-3-methylglutaryl coenzyme A reductase activities, and their competition with AA for enzymatic action and the resultant reduction in the inflammatory response (Pinsu, 2008).
Although we are able to convert dietary α-linolenic acid into eicosapentaenoic, docosapentaenoic and docosahexaenoic acids (which are found in fish and fish oils). this conversion is not efficient in people who consume a typical Western diet. Consequently, following the consumption of foods containing α-linolenic acid, our tissues are exposed to very little of the types of omega-3 fatty acids found in fish and fish oil (Surette, 2008).
The fundamental mechanism by which omega-3 FA appear to mitigate risk for CHD begins with the enrichment of membrane phospholipids with EPA and DHA. Once these long chain n-3 FA are resident in cell membranes, they may have at least four separate effects. First, because of their highly unsaturated nature, they may alter membrane properties. This can have the secondary effect of changing the microenvironment of transmembrane proteins (e.g., receptors) altering the manner in which they interact with their ligands. Altering membrane FA composition can also affect the ability of membrane-associated proteins to actually associate with the membrane and consequently to interact with other multi-protein complexes involved with cell signaling systems.
In addition, a variety of cell stressors (e.g., inflammatory mediators) interact with transmembrane receptors and subsequently initiate intracellular G-protein linked responses, one of which is the activation of phospholipase A2 (PLA2). This enzyme hydrolyzes long-chain omega-6 and omega-3 FA esterified to inner leaflet phospholipids, liberating them and making them available for conversion to a wide variety of eicosanoids via cyclo-oxygenase, lipoxygenase, and cytochrome P-450 monooxygenases.
These molecules powerfully influence cellular metabolism. PLA2-liberated omega-3 FA may directly modify the activity of ion channel themselves, resulting in altered resting membrane potentials. Finally, intracellular omega-3 FA are also able to serve as ligands for a variety of nuclear receptors (e.g., peroxisome proliferation activated receptors (PPARs), sterol receptor element binding protein (SREBP)-1c, retinoid X receptor, farnesol X receptor, and hepatocyte nuclear factor-α which impact inflammatory responses and lipid metabolism.
3. FUNCTIONAL OF OMEGA-3
Feeding humans fish oils has been shown to reduce oxygen-derived free radical formation in neutrophils and monocytes, and to enhance NO (Nitric Oxide) production by cultured human endothelial cells. Speculatively, it is also possible that a reduction in the formation of oxygen derived free radicals by endothelial cells and thus increased bioavailability of NO (Goodfellow et al., 2000). In addition, omega-3 FAs especially EPA and DHA contribute benefits through their antiarrhythmic, anti-inflammatory, antithrombotic effects. Moreover, EPA and DHA also improve vascular endothelial function and help lower blood pressure, platelet sensitivity, brain and retina (Alshatwi & Alrefai, 2007 ; Dennis et al., 1993).
Omega-3 fatty acids affect health and disease has led to a large body of evidence which suggests that these dietary lipids modulate numerous processes, including brain and visual development, inflammatory reactions, thrombosis, carcinogenesis. And also reduce mortality in non-transplanted patients. Omega-3 can reduce hypertension early after heart transplantation. Omega-3 in some reports also improve renal function in chronic progressive renal disease and kidney graft recipients (Holm et al., 2001).
This positive affect of omega-3 fatty acids was primarily associated with higher DHA levels. Previous work on omega-3 fatty acids has suggested that DHA has greater benefit than EPA for lowering blood pressure. The positive effect of higher omega-3 fatty acid levels on endothelial function raises the possibility that increased consumption of omega-3 fatty acids beginning early in life may favorably alter the risk of cardiovascular disease. Omega-3 is possible is possible improvements in endothelial function may be responsible for this reduced risk of heart disease in diabetic patients (Mori et al., 1999 ; Hu et al., 2003).
Presumably as a consequence of these cell membrane effects, omega-3 FA can diminish the activity of inflammatory cells and reduce levels of certain inflammatory mediators, which may ultimately result in reduced arterial plaque fragility. The decrease in serum triglycerides that is produced by intakes of 3-4 g/d of EPA+DHA appears to be secondary to increased hepatic beta-oxidation and decreased lipogenesis, which themselves are the result of modulation of the nuclear receptor actions (Calder et al., 2006).

Omega-3 fatty acids are being increasingly promoted as important dietary components for health and disease prevention. These fatty acids are naturally enriched in fatty fish like salmon and tuna and in fish-oil supplements. An increasing number of foods that are not traditional sources of omega-3 fatty acids, such as dairy and bakery products, are now being fortified with small amounts of these fatty acids. Fatty acids have many fates in the body, including β-oxidation for energy, storage in depot fat or incorporation into phospholipids, which form the major structural components of all cellular membranes Because humans do not have the enzymatic machinery required to synthesize omega-3 fatty acids, they must be obtained from the diet (termed “essential fatty acids”) (Suretter, 2008).
Omega-3 fatty acids are a family of fatty acids that contain two or more double bonds (“polyunsaturated”). One of which is located three carbon positions from the methyl terminus (“omega-3” or “n-3”) because the body is unable to synthesize in appreciable amounts omega-3 fatty acids that are longer than 14 carbons they are obtained primarily from dietary sources (Friedman & Moe, 2006).
Fish such as tuna, trout and salmon are especially rich sources of these fatty acids. Fish oil supplements are also a rich source as they typically contain 30%–50% omega-3 fatty acids by weight. Small quantities of omega-3 fatty acids are naturally present in meats like beef, pork and poultry. But also is plant sources such as flax and canola oil. The marine omega-3 fatty acids preparation we used contains a small amount of antioxidant vitamin E (equal to approximately 16 IU vitaminE/day), which theoretically may be expected to have an effect. (Surette, 2008 ; Goodfellow et al., 2009).
